Arabidopsis Disulfide Reductase, Trx-h2, Functions as an RNA Chaperone under Cold Stress

چکیده

The thioredoxin-h (Trx-h) family of Arabidopsis thaliana comprises cytosolic disulfide reductases. However, the physiological function Trx-h2, which contains an additional 19 amino acids at its N-terminus, remains unclear. In this study, we investigated molecular Trx-h2 both in vitro and vivo found that overexpression (Trx-h2OE) lines showed significantly longer roots than wild-type plants under cold stress. Therefore, further role Our results revealed functions as RNA chaperone by melting misfolded non-functional RNAs, facilitating their correct folding into active forms with native conformation. We binds to efficiently melts nucleic (ssDNA, dsDNA, RNA), facilitates export mRNAs from nucleus cytoplasm Moreover, increased survival rate cold-sensitive E. coli BX04 cells low temperature. Thus, our data show performs stress, thus increasing plant tolerance.

Conserved TRAM Domain Functions as an Archaeal Cold Shock Protein via RNA Chaperone Activity

Cold shock proteins (Csps) enable organisms to acclimate to and survive in cold environments and the bacterial CspA family exerts the cold protection via its RNA chaperone activity. However, most Archaea do not contain orthologs to the bacterial csp. TRAM, a conserved domain among RNA modification proteins ubiquitously distributed in organisms, occurs as an individual protein in most archaeal p...

RNA Chaperone Function of a Universal Stress Protein in Arabidopsis Confers Enhanced Cold Stress Tolerance in Plants

The physiological function of Arabidopsis thaliana universal stress protein (AtUSP) in plant has remained unclear. Thus, we report here the functional role of the Arabidopsis universal stress protein, AtUSP (At3g53990). To determine how AtUSP affects physiological responses towards cold stress, AtUSP overexpression (AtUSP OE) and T-DNA insertion knock-out (atusp, SALK_146059) mutant lines were ...

Heat-shock dependent oligomeric status alters the function of a plant-specific thioredoxin-like protein, AtTDX.

We found that Arabidopsis AtTDX, a heat-stable and plant-specific thioredoxin (Trx)-like protein, exhibits multiple functions, acting as a disulfide reductase, foldase chaperone, and holdase chaperone. The activity of AtTDX, which contains 3 tetratricopeptide repeat (TPR) domains and a Trx motif, depends on its oligomeric status. The disulfide reductase and foldase chaperone functions predomina...

Human Enterovirus Nonstructural Protein 2CATPase Functions as Both an RNA Helicase and ATP-Independent RNA Chaperone

RNA helicases and chaperones are the two major classes of RNA remodeling proteins, which function to remodel RNA structures and/or RNA-protein interactions, and are required for all aspects of RNA metabolism. Although some virus-encoded RNA helicases/chaperones have been predicted or identified, their RNA remodeling activities in vitro and functions in the viral life cycle remain largely elusiv...

Auxin response in Arabidopsis under cold stress: underlying molecular mechanisms.

To understand the mechanistic basis of cold temperature stress and the role of the auxin response, we characterized root growth and gravity response of Arabidopsis thaliana after cold stress, finding that 8 to 12 h at 4 degrees C inhibited root growth and gravity response by approximately 50%. The auxin-signaling mutants axr1 and tir1, which show a reduced gravity response, responded to cold tr...
